Straight out of 14 CFR 119.3

Domestic - Any scheduled operation conducted in: turbojet powered airplanes, airplanes having a passenger configuration of more than 9 seats, airplanes having a payload of more than 7500 pounds, between any points within the 48 contiguous states, soley within the contiguous 48 states, or specially authorized operations between any point within the contiguous 48 states and any specifically authorized point outside the contiguous 48 states.

Flag - Same as domestic, but involves operations to/from the contiguous 48 states and points outside the US (including Alaska and Hawaii).

Supplemental - Any common carriage operation for compensation or hire conducted in: airplanes with more than 30 passenger seats, airplanes with more than 7500 pounds payload capacity, prop airplanes with 9 to 30 seats that are also used in domestic or flag ops, turbojet powered airplanes with 30 seats or less that are also used in domestic or flag ops. Involves all-cargo operations, passenger carrying public charter under Part 380, or operations where the departure/arrival locations and times are negotiated with the customer (essentially, 135 on demand with big metal)

All of these operations are conducted under 14 CFR 121.
